The measured phenolic content of high phenolic E.V.O.O.s may vary greatly depending on the method of measurement. Here is a short reference to the 2 most prevalent of them: • H.P.L.C. method (with 2 variations : HPLC -Acidic Hydrolysis & HPLC-COI): This method gives the lowest results and many claim that is outdated due to it's low sensitivity. However is the only recognized method of measurement from the International Olive Council (I.O.C.). Also the Hellenic Food Authority has made the use of H.P.L.C.-COI method obligatory for all Greek olive oil companies, so that they are allowed to use the relative Health Claim (E.U. 432/2012) by European Food Safety Authority (E.F.S.A.). Additionally this method has high availability as there are many international certified labs available for cross examining the results, something that increases significantly the reliability and objectivity of the method. • N.M.R. method: is a relatively new method which usually gives the higher results. It thought to measure a wider range of olive oil phenolic compounds. Many claim this to be a better method and they prefer to use it as usually gives much higher measurements (from 30% up to 500% higher than HPLC) therefore is better for marketing purposes too. However, as far as we know, there is no official recognition from any significant international organizations and it seems that it produces inconsistent results, even on the same olive oil samples. Additionally only one lab worldwide uses this method, so there is no possibility of cross examinations and therefore highly reduces it's relative objectivity & reliability. Much bigger availability and further evolution, will be probably be the key factor for a potential substitution of the HPLC method in the future . Based on the facts above, we are making periodically measurements by using all methods of measurements (including others not mentioned) and use average values in order to ensure the top quality of our product, but we have to use HPLC method as method of reference. Coming back to your original question the relative wide range of polyphenols consist of the measurements of both methods (lower values HPLC-COI, higher values HPLC-Acidic Hydrolysis & N.M.R). Olive oil (and in particular its phenolic compounds), is sensitive to light (mainly ultraviolet irradiation), air contact (oxygen) and to high temperatures, because it can be oxidized and lose part of its high-quality and valuable health protective properties. The use of glass containers with small capacity is crucial, in order to avoid oxidation from the extensive air contact. Larger glass containers, means longer-extended consuming period, and therefore extended required storing period of bottles that have been already opened. Something that causes a more extensive oxidation from air contact with larger quantity of oxygen, for a longer time period.
