How much weight can this backpack hold?

Maybe max 20 lbs? It's got good capacity, but not the strength of construction to withstand more. Not really meant for hauling gear...

Is it just one big open space inside, or are there pouches/pockets?

Their are 2 pouches/pockets in the main area. Another thin zippered area in front of that with 2 mesh pockets. Another thin area in that vertical zipper on the front. Also, a mesh pocket on each side.

How secure is backpack to prevent theft?

other than the obvious front middle zipper - I felt it was secure. There was an inner pocket that I put my passport, cards and cash in. You should note that I returned this backpack. I used it for a very strenuous hike and got very sweaty. In side the pack I carried two white shirts at the bottom of the main compartment. I had a yellow pack and the yellow die leached onto both shirts during my hike. It was very hot and I was very sweaty - but the color should not have leaked.
