Hello, can anyone tell me is there a major difference, between a usb 3.0 and a usb 2.0?  thank you.

usb 3 is an upgrade on the older usb 2 standard.  usb 3 runs much faster in part because its cable has more wires in it.  usb 3 uses a blue connector base pad inside of the plug and the older usb 2 normally use black or white.  This cable extends the usb 2 port on the side of your computer to some point 9 feet away.  usb 3 equipment is built to operate with a usb 2 port at the old slower speeds.  If your computer has only usb 2 ports this cable will be as good as you will get.  If you are using a usb 2 device this cable is as good as you will get.  If your computer has usb 3 ports and you are using an usb 3 device this cable will result in a large speed loss.

I'm not familiar with the electronic needs of i-devices, whether they require specific cords. This is just a simple USB extension cord, with a male USB tip on one end, and a female USB tip on the other, so you can plug your (male ended) USB charging cord from any device into it. I got mine because I didn't like the 2 foot cord provided with one of my electronic devices, and I have absolutely no complaints.  I've not had any problems at all.  That said, all my devices are Android.

Will this work to make a usb 2.0 work in usb 3.0?

Your USB 2.0 device will still operate at the same speed when you connect to a USB 3.0 port or extension cord. It will not magically speed up. The USB 3.0 interfaces have more wires to support the increased speed. Your existing USB 2.0 does not have these wires.

if i connect my 3.0 usb thumb drive into a 2.0 ext. cable then plug into my 3.0 laptop port.  will i lose transfer speed because of the 2.0 cable

You will, yes. USB 3 is backwards compatible with 2.0, but there is a different connection that USB 3 ports use that NEED USB 3 at all steps in the connection process to properly utilizae.
