No, you don't. If you don't, it will drain and filter a bit. To get a stronger suction for filtering, you have to keep squeezing the bulb.

Can you use this in reverse to fill the tank with water?

You can not hook it up to a faucet, but you could from a bucket. But the bucket has to be higher then the tank. It still works on gravity

How can I request a replacement. Small one way plastic valve had fallen out of position on first use. Won't work at all without it.

A replacement? If you just buy it I would recommend to just refund it and get a new one.ones the valve stopped working is not the same,just get new one

Would this work for cleaning an aero garden?

I don’t have an aero garden but I don’t see why it wouldn’t work. The basic need is the same.
