You shouldn’t have no pressure loss. You shouldn’t leak test for a few hours or overnight. 30 minute leak test with zero pressure loss your 100% to go to fill with liquid.

How are the 0.05 bar increments measured on the guage? Is it every 2 lines or 1 line?

every line is around 0.05 bar, I say around since you have nine lines between 0 to 0.25 and 10 lines between 0.25 to 0.5

Will this work when putting all tubes together then doing the leak test on the fill port? I read someone was doing it individually how??

The way you are thinking is by building your whole loop, while that will work, if you do end up having a leak it'll be a pain to find it. Testing individually would by pumping up each block/pump/reservoir/radiator individually as to isolate and possible leaks immediately. If you use the one by one method I'd conduct another test once its all built.
