Malt Extract Agar (perfect for mushroom tissue cultures). A blend of high quality Malt Extract, Peptone G, Dextrose and cleanly processed Base Agar makes this an excellent mix for mycelium growth. 100 grams will yield approximately 1.8 Liters of MEA Solution which will yield 70-80, 100mm Petri Dishes. FORMULA: 35.7% Malt Extract 1.8% Pancreatic Digest of Gelatin (Peptone G) 35.7% Dextrose 26.8% Agar Agar SOLUTION INSTRUCTIONS: Step 1: Dissolve 100g of Malt Extract Agar (MEA) in 1.8 Liters of Distilled Water Step 2: Bring to a Boil and Stir Step 3: Autoclave at 121 degrees Celsius for 15 minutes Step 4: Pour into Petri Dishes

Review: Excellent Agar - Modified Smaller Use Recipe Included In My Review - FANTASTIC and economical for my Brewery Laboratory for plating post packaged products to check for Microbiological stability. Very easy to prepare and keeps extremely well. I have modified the "recipe" to use for a smaller quantity of plates: Weigh out exactly 16.7 grams of MEA. Fill flask with 300 mL of water and add 1 stir bar. Place flask on magnetic heating stir plate and set heat to 8. Set stirring rate slowly to a maximum speed of 5 to create a medium vortex. Add MEA to 300 mL of water in flask. Stir until completely dissolved and bring to a boil. The agar is then poured into heat proof bottle(s) filled no further than 2/3. The lid of the bottle is to be closed fully, then released ¼ turn prior to sterilizing. Sterilize the bottle using laboratory microwave for 5 minutes on full power. Allow the agar to cool to approximately 120 degrees Fahrenheit, checking frequently with temperature gun. Thoroughly wash and dry hands and put a fresh pair of nitrile gloves on. Create a sterile area by thoroughly wiping the bench with 70% alcohol and paper towel. Do not completely dry the alcohol, allow it to evaporate. Air conditioner must be turned off, doors kept closed and foot traffic restricted prior to and at time of pouring. Organize petri dishes base down (the smaller section of the two halves) on the bench. Hold the flask containing sterile agar medium in liquid form with your right hand. Turn your left hand palm side up and clamp the bottle lid between two fingers. Use the hand holding the lid to lift each lid of the Petri dishes. Pour approximately 15-20 mL of the agar medium into the base of each Petri dish until approximately half full. Hold each petri dish lid so it partially covers the bottom of the dish as you pour. Immediately place the lid on the base at an angle so steam is able to escape. Allow the plates to cool to room temperature for approximately 30 minutes before packing upside down into labelled plastic bags, or use the bags the plates came in. Refrigerate the plates, storing for approximately 30 days maximum prior to use.

## Questions & Answers

**Q: If you do not have an autoclave available but otherwise have a sterile area for inoculation would it still work? Does the agar have to be autoclaved?**
A: a pressure cooker works really well in place of an autoclave, I use one myself. 15psi for various amounts of time depending on the material.  about 45 min for Malt Extract Agar in a 500ml or 1000ml media specimen bottle (with a proper autoclavable lid, left slightly loosened to allow pressure release from the internal expanding boiling gases without letting in steam and changing the water balance of the mix as well.. basically just enough to be a pressure valve or it may shatter.. from internal pressure.) works great. I pour the dishes in a still air box that I set up with HEPA filtration ventilation for positive air flow as a makeshift laminar flow hood using positive sterile air flow to keep the germs and molds and whatnot from my dishes when I pour, tho the heat does a good job as well, as you want to pour while it's still fairly warm (around 150f).. my contamination rate is super low, 0% on my last batch of 120 dishes in fact but never has been higher than 4% (basically 4 dishes out of 100 showing any kind of growth on them a week after initial pour). I also wear gloves and use 70% iso alc for disinfecting into the box.

**Q: I would like to know how many grams of mea per 100ml would i need to make culture slants for brewers yeast? thanks**
A: 100g/1800ml will give g per ml if you need smalller increments.

**Q: Is it really necessary to boil before autoclaving? other mea's ive used you just mix it in your vessel and autoclave for 45mins. would this be ok?**
A: You want to mix it with boiling/hot water so it gets mixed evenly

I have poured this in an sab and worked great. For small batches I used 1g per 18 ml of water. I then used a “no pour tek” which sterilized the agar in the container and worked well for that as well.
